Histone-lysine N-methyltransferase, H3 lysine-79 specific

Dot1L mediates H3K79 methylation in vivo and in vitro. It is a non-SET domain containing methyltransferase and its active site binds to S-adenosyl-L-methionine (SAM) via SAM binding pocket (PMID: 12628190). Dot1L is reported to form a fusion protein with MLL (mixed lineage leukaemia) causing upregulation of leukaemia associated genes thus promoting leukaemic transformations (PMID: 15851025). Dot1L levels are significantly reduced in patients suffering from dilated cardiomyopathy (DCM) suggesting its importance in many heart related abnormalities (PMID: 21289070)
