Abstract:
Most interestingly, this entire pathway is controlled by insulin, and therefore responds to fed/fasted states of the animal. Insulin signalling activates phospholipase-D to generate phosphatidic acid on LDs, which signals on LDs to recruit kinesin-1 motors, consequently transporting reactive LDs to the smooth-ER inside hepatocytes. Our results provide multiple molecular players for targeted intervention against hyperlipidemia and fatty liver. This possibility is demonstrated using a specific peptide that removes kinesin-1 from LDs and diminishes triglyceride secretion with no apparent effect on cell viability.
