E3 ubiquitin-protein ligase RING2

RING1B mediates ubiquitination of H2A lysine 119. It is part of a human polycomb repressive complex 1-like (hPRC1L) complex that consists of Ring1, Ring2, Bmi1 and HPH2 (PMID: 15386022). Mel-18 is known to guide RING1B to its substrate (PMID: 17936708). USP7 is reported to modulate the activity of RING1B by deubiquitinating and stabilizing it (PMID: 20800574). RING1B activity seems to be involved in the compaction of DNA and formation of higher order chromatin structure which causes the repression of Hox genes and this function is independent of the histone ubiquitination (PMID: 20471950). It is shown to be phosphorylated by growth factors and signalling molecules like p38 MAPK and ERK1/2 pathways (PMID: 19405034). Ring1b is also responsible for ubiquitination of lysine 121 on H2A.Z. This monoubiquitinylated H2A.Z is associated with inactive X chromosome (PMID: 17636032).
