E3 ubiquitin-protein ligase UHRF1
NP95 is reported to show in vitro ubiquitination of histone H3 (PMID: 14993289). Loss of NP95 leads to impairment of histone deacteylation that is important for hetrochromatinisation and hints at the HDAC recruitment activity of NP95 (PMID: 17182844). It has been shown to bind methylated H3K9 and mediate ubiquitination of H3 that helps in proper heterochromatin formation (PMID: 17967883). Recent evidences suggest that NP95 is involved in the cross-talk that exists between methylated DNA and histone modifications (PMID: 19077538)(PMID: 20026581). It also seems to interact with the histone acetyltransferase Tip60 (PMID: 19800870). NP95 is involved in sensitisation of breast cancers by inhibition of MDR1 (PMID: 20037778).
