E3 ubiquitin-protein ligase RNF168

RNF168 belongs to RNF168 family. Ubiquitinates histone H2A in vitro and in vivo and is responsible for creating 'lysine-63' linked polyubiquitin chains (PMID: 19500350). During DNA damage RNF168 is recruited to the damage foci and helps in recruitment of other factors. Loss of this activity is implicated in RIDDLE syndrome(PMID: 19203578). Recently it has been shown that histone ubiquitination activity of RNF168 is also involved in the transcriptional silencing of DNA regions associated with double strand breaks during DNA damage (PMID: 20550933).
