Lysine-specific demethylase 5B
PLU-1/JARID1B belongs to the JARID1 family of demethylases that catalyse the demethylation of H3K4me3. This activity is responsible for removal of activation mark and repression of tumour suppressor genes which leads to proliferation of breast cancers (PMID: 17363312). It is also involved in cell cycle progression form G2 to M phase and E2F1,E2F2 are shown to be downstream targets in PLU-1 pathway (PMID: 17709396, PMID: 20226085). Expression levels of PLU-1 is reported to be significantly altered during melanoma development and in prostate cancers (PMID: 17973255, PMID: 18048344). PLU-1 also regulates the androgen receptor mediated transcriptional activation (PMID: 18048344). Using JARID1B as a biomarker, a subpopulation of melanoma cells have been identified that have a doubling time of more than 4 weeks and contributes to the maintenance of tumours. It has been proposed that dynamic expression of JARID1B does not follow the conventional cancer stem cell model (PMID: 20478252).
