Lysine-specific histone demethylase 1A
LSD1 is a lysine specific histone demethylase that belongs to the flavin monoamine oxidase family of flavin containing proteins and demethylates mono and dimethylated H3K4/K9 (PMID: 15620353, PMID: 16079795, PMID: 15811342). It is part of a complex called REST (RE1-silencing transcription factor) and its recruitment by a REST corepressor CoREST enables LSD1 to demethylate nucleosomal H3K4me2 and mediate silencing of REST target genes (PMID: 16079794). LSD1 is reported to cooperate with JMJD2C demethylase to complete demethylation of H3K9me3 repressive marks so as to facilitate androgen receptor mediated transcriptional activation (PMID: 16079795, PMID: 17277772). It is also shown to be negatively regulated by BHC80 (PMID: 16140033). LSD1 controls the demethylation of p53 molecule and thereby negatively regulates its pro-apoptotic function (PMID: 17805299). Oestrogen-dependent transcriptional activation requires LSD1 dependent demethylation of target gene promoters which produces local hydrogen peroxide required for recruitment of other regulatory factors (PMID: 18187655). LSD1 is heavily implicated in carcinogenesis. Levels of LSD1 are reported to be significantly upregulated in ER negative breast cancers and can serve as a biomarker for diagnosis (PMID: 20042638). LSD1 is shown to be involved in the proper segregation of mitotic chromosomes (PMID: 20189264).
