CREB-binding protein
The acetyltrasferase activity of CBP is shown to be directly involved in transcriptional activation of genes (PMID: 9582282). CBP often acts in conjugation with p300. CBP/p300 associates with other histone acetyltransferases like PCAF. There are about 400 proteins that interact with CBP/p300 (PMID: 20110770). Mutations in CBP (some in the conserved HAT domain) are implicated in Rubinstein-Taybi Syndrome (RTS) (PMID: 15706485). RNA interference mediated knockdown of CBP and p300 in immortal HeLa cells results in cell death due to "mitotic catastrophe" and "chromosome shredding"(PMID: 17272271). Artificial enhancement of the HAT activity of CBP via Phenylephrine has been shown to induce cardiac hypertrophy (PMID: 12477714). Acetylation of different lysine residues of histone H3 and H4 by CBP/p300 at the site of double stranded DNA breaks generated during IR radiation have been shown to recruite SWI/SNF chromatin remodellers for DNA repair. Hence, inhibition of CBP activity can help is sensitising cancer cells to radiotherapy or chemotherapy (PMID: 21217779).
