Histone-lysine N-methyltransferase SETD8
PR-SET7 contains a SET domain and whether it is a monomethylase or a dimethylase is debatable. Some reports state that it is exclusively a monomethylase (PMID: 15933069) whereas others show a switch from mono to dimethylatation at lysine 20 position of histone H4 upon PR-SET7 activation (PMID: 15200950). PR-SET7 mediated H4K20 methylation status (mono- or di-) contributes to chromosome behaviour during mitosis and is required for proper cytokinesis (PMID: 15200950). PR-SET7/SET8 is necessary for progression of cell cycle to S phase (PMID: 18166648). H4K20 monomethylation helps in loading of the pre-RC proteins on the origin of replication and plays an important role during initiation of DNA replication (PMID: 20953199). PR-SET7 is shown to be undetectable during S phase and its interaction-mediated degradation via PCNA in context of DNA damage is required for maintaining the integrity of the genome (PMID: 21035370).
